IntheNeuroProbeAC48chemotaxischamber,chemoattractantsaddedtothelowerwellsactoncellsinacellsUSPensionaddedtotheupperwells.Apairoffiltersseparatestheupperandlowerwells.Afterincubationthemigratedcellsonorinthefiltersarestained.Withcellulosenitratefilters,onemeasuresthedepththecellshavereachedwithinthefiltermaterial;withpolycarbonatefilters,onecountsthecellsthathavemigratedthroughthefilterandadheredtoitsunderside.TheAC48hasspecialfilter-retaininghardwaredesignedtoholdthetwofiltersinplaceduringassemblyanddisassemblyofthechamber.
TheAC48consistsofprecision-machinedacrylictopandbottomplates,assemblyhardware,andasiliconegasket.Thebottomplatehas48wells,eachwith25µLvolumeandadiameterof3.2mm.Correspondingholesinthetopplateformtheupperwellswhenthechamberisassembled.Thechamberfeatureseightbutton-headscrewspermanentlymountedinthetopplate,whichholdthegasketandfiltersinplaceduringassemblyanddisassembly.Thisfilter-retaininghardwareispositionedtoaccommodatetwo35mmx41mmpiecesoffiltermaterial,eachcovering24wells.Narrowraisedrimsaroundmatingsurfacesofthetopandbottomwellsenhancetheseal.
TheAC48isdesignedespeciallyforusewithcellulosenitratefilters.Polycarbonatefiltersmayalsobeused,butifthatisyour primary interest,wesuggestyoualsoconsiderthemorecommonlyusedAP48chamber.
Includedwitheachchamberisan AC48protocol detailingproperuseandcareofthechamber,aswellasvariousprotocolsfor stainingcellulosenitratefilters.
