TheNeuroProbeDC8cellsettlingchamberisdesignedtodepositcellsinamonolayeronaglassmicroscopeslide.Afterthewickingandsettlingprocessinthechamber,cellsadheretotheslideinarandomdistributionsuitablefordifferentialcounting,withexcellentcellularmorphology.
Thechamberconsistsoftwoprecision-machinedacrylicplates,asiliconegasket,andassemblyhardware.Thelowerplatehasarecesstofita25x75mmglassmicroscopeslide.Theupperplatehaseightthroughholes,whichform280µLwellswhentheupperplateisclampedontothemicroscopeslideinthelowerplate.Thegasketandasheetofprepunchedfilterpaper(Stock#DC8WF1)pinchedbetweentheplatescauseliquidinthecellsUSPensiontowickawayasthecellssettleontotheslide,leavingthecellsitesdry.
Forbestresultsmicroscopeslideswithenhancedbindingpropertiesandahydrophobicmaskaroundtheeightsettlingsites(Stock#DC8SC)areavailable.TheseslidesimprovecelladherenceandalsorestrictPipettedsolutionstoindividualsites.Differentsolutionscanthenbeappliedtosettledcellswithoutcrosstalk.
Whenworkingwithcellsofrelativelylowdensity,suchaslymphocytes,thechambershouldbespuninacentrifugeequippedwithamicroplatecarrier.Acentrifugeaccessorypack(Stock#DC8AP)providesanadapterandcounterweightforthispurpose.
