IntheNeuroProbeAP48chemotaxischamber,chemoattractantsaddedtothelowerwellsactoncellsinasUSPensionaddedtotheupperwells.Afilterseparatestheupperandlowerwells.Afterincubationthemigratedadherentcellsontheundersideofthefilterarestainedandcounted.
TheAP48consistsofprecision-machinedacrylictopandbottomplates,assemblyhardware,andasiliconegasket.Thebottomplatehas48wells,eachwith25µLvolumeandadiameterof3.2mm.Correspondingholesinthetopplateformtheupperwellswhenthechamberisassembled.Asingle25x80mmpieceoffiltermembraneisplacedbetweenthetopandbottomplates,andagasketispositionedoverthefiltertocreateaseal.Narrowraisedrimsaroundmatingsurfacesoftopandbottomwellsenhancetheseal.
Beforethemigratedcellscanbestainedandcounted,thenonmigratedcellsmustbewipedoffthetopsideofthefilter.An accessorypack(stock#P48AP)includesinstrumentsforthispurpose.
TheAP48isdesignedforusewithpolycarbonateandcellulosenitratefilters.Pleasenotethatwealsosellthe AC48,a48-wellchamberspeciallydesignedforusewithcellulosenitratefilters
