IntheNeuroProbeA3BP48chemotaxischamber,chemoattractantsaddedtothelowerwellsactoncellsinasUSPensionaddedtotheupperwells.Thelowerwellsareformedbyclampingthemiddleplatetothebottomplate.Afilterseparatestheupperandlowerwells.Afterincubationthemigratedadherentcellsonthisfilterarestainedandcounted,asinthewidelyused AP48.Inaddition,amembranecanbeplacedonthebottomofthebottomwellstocollectmigratednonadherentcellstobestainedandcounted.Ifthereisnomembraneinthelowerwells,nonadherentmigratedcellscanbeviewedthroughtheclear5.7mmthickbottomplateoftheA3BP48withaninvertedmicroscope.
TheNeuroProbethree-tieredchamberhasthreeprecision-machinedacrylicplates,assemblyhardware,andtwosiliconegaskets.Theclearflatbottomplatehashardwareholesonly.Themiddleplatehas48throughholesandsixpermanentlyinstalledhardwareposts.Thetopplatehashardwareholesand48throughholes.Raisedrimsaroundtheholesofthemiddleplateformsealsonbothsideswhenitandthetwogasketsareclampedbetweenthetopandbottomplates.
Beforethemigratedcellsadheringtothemiddlefiltercanbestainedandcounted,thenonmigratedcellsmustbewipedoffthetopsideofthefilter.Anaccessorypack (Stock#P48AP) includesinstrumentsforthispurpose.
TheA3BP48isdesignedforusewithpolycarbonatefilters,althoughcellulosenitratefilterscanbealsobeused.
Accompanyingeachchamberisan A3BP48protocol foritsuseandcare.
